      为探究延黄牛PLK2基因单核苷酸多态性及其与肉质性状的相关性,研究以96头延黄牛为研究对象,利用PCR测序方法检测PLK2基因多态性,分析基因多态性与肌肉氨基酸、脂肪酸含量的连锁关系。结果显示,延黄牛PLK2基因第六外显子存在1个突变位点(Chr20:20,849,922C>T),产生三种基因型,CC为优势基因型。该位点基因纯合度、有效等位基因数分别为0.635、1.573;多态信息含量为0.298,属中度多态。连锁分析发现CC基因型个体的缬氨酸含量显著大于CT型。CC、CT基因型个体的棕榈酸、棕榈油酸极显著高于TT型,油酸含量显著高于TT型;TT基因型个体的亚油酸含量极显著高于CC、CT型,硬脂酸、花生四烯酸含量显著高于CC、CT型。初步证明,PLK2基因可作为延黄牛肉质性状选育提高的潜在分子标记。

      In order to explore the single nucleotide polymorphism of PLK2 gene and its correlation with meat quality traits of Yanhuang cattle, 96 Yanhuang cattle were selected as the research objects, and the PLK2 gene polymorphism was detected by PCR sequencing. The linkage relationship between gene polymorphism and muscle amino acid and fatty acid content was analyzed. The results showed that there was a mutation site (Chr20:20,849,922C>T) in the sixth exon of PLK2 gene of Yanhuang cattle, and three genotypes were produced, CC being the dominant genotype. The homozygosity and effective allel numbers of this locus were 0.635 and 1.573, respectively. The polymorphism information content was 0.298, which belonged to moderate polymorphism. Linkage analysis showed that the valine content of CC genotype was significantly higher than that of CT genotype. The contents of palmitic acid and palmitoleic acid in CC and CT genotypes were significantly higher than those in TT genotype, and oleic acid content was significantly higher than that in TT genotype. The content of linoleic acid in TT genotype was significantly higher than that in CC and CT genotype, and the content of stearic acid and arachidonic acid was significantly higher than that in CC and CT genotype. It was preliminarily proved that PLK2 gene could be used as a potential molecular marker for the improvement of meat quality traits in Yanhuang cattle.
