[目的]旨在探讨SIRT3基因多态与德州驴生长性状之间的关系,寻找与不同性别的德州驴体尺性状相关的分子标记。[方法]本研究采用质谱分型方法检测190头健康成年德州母驴及214头德州公驴SIRT3基因的多态性,分析其多态位点不同的基因型与不同性别德州驴生长性状的关联性。[结果]结果发现,SIRT3基因扩增序列区间存在2个SNPs位点,分别位于3’-UTR区域和外显子区域。在SIRT3-1(g.19763T>C)位点,公、母德州驴的优势等位基因型均为TC,C是SIRT3-1位点的优势基因,公驴和母驴TT型个体均值在体高和尻长方面均显著高于TC型个体均值(P <0.05),公驴中TT型个体均值在尻高方面显著高于TC型个体均值(P<0.05),而CC型个体均值在管围方面显著高于TT型个体均值(P<0.05)。在SIRT3-2(g.35148A>G)位点,公母德州驴的优势等位基因型均为GG,G是优势基因,公驴和母驴GG基因型个体均值在胸宽方面显著高于AG型个体均值(P<0.05),公驴GG型个体均值在尻宽方面极显著高于AG型个体均值(P<0.01)。[结论]综上,可以将SIRT3基因多态作为影响德州驴生长性状的候选基因用于标记辅助选择,为德州驴的选育工作提供科学依据。 |

[Objective]The purpose of this study was to explore the possibility of SIRT3 gene as a candidate gene for growth traits of Dezhou donkeys and to find molecular markers related to the body size traits of different genders of Dezhou donkeys. [Methods]In this study, the polymorphism of SIRT3 gene in 190 healthy adult Dezhou Jennis and 214 Dezhou Jackasses was detected by mass spectrometry, and the association between different genotype inviduals in polymorphic loci and growth traits in Dezhou donkeys of different genders was analyzed. [Results]The results showed that there were two SNPs loci located in the 3''-UTR and exon region in the SIRT3 gene. At SIRT3-1 (g.19763T > C), TC was the dominant genotype in both Jackasses and Jennis, and C was the dominant allele. The mean value of TT type individuals in Jackasses and Jennis was significantly higher than that of TC type individuals in terms of body height and rump length (P < 0.05). In Jackasses, the mean value of TT type individuals was significantly higher than that of TC type individuals in terms of rump height (P < 0.05), while the mean value of CC type individuals was significantly higher than that of TT type individuals in terms of cannon circumference (P < 0.05). At SIRT3-2 (g.35148 A > G) locus, the dominant genotype of Jackasses and Jennis was GG, and G was the dominant allele. The average chest width of GG genotype of Jackasses and Jennis was significantly higher than that of AG genotype (P < 0.05), and the average rump width of GG genotype of Jackasses was significantly higher than that of AG genotype (P < 0.01). [Conclusion]In conclusion, SIRT3 gene can be used as a candidate gene for marker-assisted selection of Dezhou donkeys, which provide scientific basis for the breeding of Dezhou donkeys. |
