      [目的]以云南省迪庆州香格里拉市高山放牧和昆明市东郊小哨示范牧场圈养育肥1.5年以上的成年中甸牦牛为研究对象,比较两种饲养条件下组织器官中HIF-1α基因mRNA表达差异。[方法]采用实时荧光定量PCR(RT-PCR)技术检测HIF-1α基因在不同饲养条件下中甸牦牛不同组织中的表达水平。[结果]结果表明:中甸牦牛不同器官组织HIF-1α基因的相对表达量差异极显著(P<0.01);在不同饲养条件下相对表达量差异显著,高山放牧显著高于异地圈养育肥(P<0.05);在高山放牧的中甸牦牛肝脏中相对表达量显著高于圈养育肥的(P<0.05)。[结论]中甸牦牛不同组织器官中HIF-1α基因广泛表达且具有组织特异性,在不同海拔饲养的相对表达量具有显著差异,为牦牛的耐低氧机制研究提供了理论依据。

      [Objective]In this study, adult Zhongdian yaks grazing on alpine pasture in Shangri-La of Diqing prefecture, and fattening >1.5 years in captivity on the eastern suburb of Kunming, were used to reveal the expression differences of HIF-1α gene from different tissues under two different feeding conditions. [Methods] Real time PCR (RT-PCR) was used to determine the expression levels of the HIF-1α gene in different tissues in Zhongdian yak under different feeding conditions. [Result] The results showed that: the relative expression of the HIF-1α gene in Zhongdian yak varied significantly in different tissues (P<0.01), and under different feeding conditions, it was significantly higher in Alpine grazing than that the captive animals (P<0.05), and it was significantly higher in liver from alpine grazing than that in captive animals (P<0.05). [Conclusion] HIF-1α gene was widely expressed in different tissues in Zhongdian yak, with significant tissue-specificity. There were significant differences of relative expression of HIF-1α from different altitudes, providing a theoretical basis for the mechanism of hypoxia tolerance in yaks.
