      探究长链非编码 RNA(long non-coding RNA, lncRNA)在牛肌内脂肪沉积中的重要作用及相关分子机制。利用转录组测序技术对 A2 和 A4 等级郏县红牛背最长肌肌内脂肪组织进行转录组测序,筛选差异表达的 lncRNA,并对其靶基因进行预测以及 GO 和 KEGG 富集分析。结果显示,在 A2 和 A4 组样本中筛选到 190 个显著差异表达的 lncRNA,其中与 A2 组相比,有 94 个 lncRNA 的表达在 A4 组中显著上调,96 个 lncRNA 显著下调。这些差异表达 lncRNA 分别顺式和反式调控 7 个和 175 个靶基因。富集分析发现,反式靶基因显著富集到脂质代谢、脂肪酸代谢以及丙酸酯代谢等通路(P≤0.05)。上述结果表明,差异表达 lncRNA 可能通过反式调控靶基因参与牛肌内脂肪沉积的调节。

      The purpose of this study was to explore the important role of long non-coding RNA (lncRNA) in the deposition of intramuscular fat (IMF) in bovine and related molecular mechanisms. Transcriptome sequencing technology was used to analyze the longissimus dorsi muscle samples of A2 and A4 grade Jiaxian Red Cattle, and the differentially expressed lncRNAs were screened, and their target genes were predicted and GO and KEGG enrichment were analyzed. The results showed that 190 differentially expressed lncRNAs were screened in the A2 and A4 groups, of which 94 lncRNAs were significantly up-regulated and 96 lncRNAs were significantly down-regulated in the A4 group compared with the A2 group. These differentially expressed lncRNAs regulate 7 and 175 target genes respectively through cis- and trans-regulation. Enrichment analysis showed that the trans target genes were significantly enriched in lipid metabolism, fatty acid metabolism and propionate metabolism pathways (P≤0.05). These results suggest that differentially expressed lncRNAs may participate in the regulation of bovine intramuscular fat deposition through trans-regulatory target genes.
